Hexapeptide-9 — Collagen-Stimulating Peptide
Hexapeptide-9 is a targeted collagen-stimulating peptide that works directly at the dermal matrix level — signalling fibroblasts to upregulate collagen synthesis and improve the structural integrity of ageing or compromised skin. First introduced to cosmetic science in 2010, it represents the more sophisticated second generation of collagen-stimulating peptides, working through a distinct signalling pathway from the well-known Matrixyl family and delivering complementary rather than redundant activity when the two are combined in the same formula.
The mechanism is precise: Hexapeptide-9 binds to receptors on dermal fibroblasts and triggers the production of both Type I and Type III collagen — the two primary structural collagens responsible for skin firmness, thickness, and resistance to mechanical stress. As collagen levels decline with age (at approximately 1% per year from the mid-twenties), the skin loses its ability to spring back, fine lines deepen into wrinkles, and the overall architecture of the face begins to deflate. Hexapeptide-9 directly addresses this by restoring the biological signal that fibroblasts need to keep producing the collagen the skin is no longer making on its own.
In formulation, Hexapeptide-9 is a clean, water-soluble active that incorporates easily into serums, moisturisers, eye creams, and treatment concentrates. It is stable, compatible with most cosmetic systems, and carries no sensory or aesthetic challenges — no colour, no significant odour, and no impact on product texture. At the recommended use level of 3–7%, it delivers meaningful activity without inflating formula cost unnecessarily. It pairs particularly well with Palmitoyl Pentapeptide-4 (Matrixyl) and Palmitoyl Tripeptide-1 for a multi-pathway collagen synthesis approach, and with Copper Tripeptide-1 (GHK-Cu) for formulators who want to combine collagen stimulation with regenerative activity in the same product.
For brands building anti-ageing claims, Hexapeptide-9 is a strong supporting ingredient that adds genuine mechanistic depth to a peptide formula. It is particularly well suited to firming serums, neck and décolletage treatments, and any product targeting loss of elasticity and structural volume in mature skin.
Formulator's notes: Add to the water phase at or below 40°C. Recommended use level 3–7%. pH stability range 4.5–7.5. Water-soluble — no special solubilisation required. Compatible with standard preservation systems, humectants, and other peptide actives. Store cool and dark; use within 12 months of opening. INCI: Hexapeptide-9.
